o-succinylbenzoic acid synthase

E coli enzyme; with SHCHC synthase catalyzes formation of o-succinylbenzoic acid from isochorismic acid & alpha-ketoglutarate in presence of thiamine pyropyrophosphate; succinic semialdehyde anion is formed by decarboxylation of alpha-ketoglutarate by thiamine pyrophosphate; anion then attacks isochorismic acid, initiating aromatization with elimination of pyruvate & H2O
Also Known As:
2-succinylbenzoic acid synthase; MenC protein, E coli; OSB synthase; o-succinylbenzoate synthase; ortho-succinylbenzoic acid synthase

05/18/2004 - "This phenomenon has been suggested to occur in the o-succinylbenzoate synthase (OSBS) from a species of Amycolatopsis that catalyzes not only the physiological syn-dehydration reaction of 2-succinyl-6-hydroxy-2,4-cyclohexadiene-1-carboxylate but also an accidental racemization of N-acylamino acids [Palmer, D. 08/01/1993 - "The benzenoid aromatic compound o-succinylbenzoic acid is formed by dehydration of the prearomatic compound 2-succinyl-6-hydroxy-2,4-cyclohexadiene-1-carboxylic acid by the enzyme o-succinylbenzoate synthase, encoded by the menC gene. "
01/13/2004 - "o-Succinylbenzoate synthase (OSBS) from Amycolatopsis, a member of the enolase superfamily, catalyzes the Mn2+-dependent exergonic dehydration of 2-succinyl-6R-hydroxy-2,4-cyclohexadiene-1R-carboxylate (SHCHC) to 4-(2'-carboxylphenyl)-4-oxobutyrate (o-succinylbenzoate or OSB) in the menaquinone biosynthetic pathway.
